RESUMO

Nucleated erythrocytes of healthy domestic chicken and ducks, and lymphocytes of healthy Sprague Dawley rats were evaluated for nucleic acid distribution pattern, employing light and fluorescence microscopy procedures, as well as digital imaging analytical methods. The results demonstrate a unique organization of nuclear DNA of mature chicken and duck erythrocytes, as well as immature duck erythrocytes, as delineated spherical nuclear bodies that mostly corresponded with euchromatin zones of the cells in routine Wright-stain blood smears. The nuclear DNA of the rat lymphocytes, on the other hand, was observed as a more diffuse green fluorescing nuclear areas, with punctate variably-sized diffuse areas of RNA red fluorescence. RNA red color fluorescence was also evident in the narrow cytoplasm of the lymphocytes, especially in large lymphocytes, in comparison with the cytoplasm of the mature avian erythrocytes that completely lacked any nucleic acid fluorescence. Nuclear RNA fluorescence was lacking in the mature chicken erythrocytes, compared with those of the mature and immature duck erythrocytes as well as lymphocytes of both avian and rats blood. The significance of these findings lies in the establishment of normal benchmarks for the nuclear and cytoplasmic nucleic acid pattern in eukaryotic cells. These normal benchmarks become valuable in rapid diagnostic situations associated with pathologies, such as the presence of viral nuclear and cytoplasmic inclusion bodies that can alter the nucleic acid pattern of the host cells, and in conditions of cellular abnormal protein aggregations. Variability of cellular nucleic acid pattern can also aid in prognostic assessments of neoplastic conditions.

OBJECTIVE: Morinda citrifolia L. is a traditional Polynesian medicinal plant which is apparently useful for bowel disorders, skin inflammation, infection, mouth ulcers and wound healing. This study aimed to evaluate the wound-healing activity of Morinda citrifolia fruit juice in streptozotocin-induced diabetic rats. METHOD: An excision wound model was used. The animals were weight-matched and placed into three groups (n = 6 per group). Group 1 animals served as normal controls, while animals in groups 2 and 3 served as diabetic controls and experimental diabetic animals respectively. All animals were anaesthetised and a full-thickness excision wound (circular area of 300 mm2 and 2 mm deep) was created. Group 3 animals were given the juice of Morinda citrifolia fruit (100 ml per kilogram of body weight) in their drinking water for 10 days. Wound area measurements were taken on days 1, 5 and 11. Blood samples were collected simultaneously for glucose measurement. Granulation tissue that had formed on the wound was excised on day 11 and processed for histological and biochemical analysis. RESULTS: The wound area of the Morinda citrifolia-treated group reduced by 73% (p < 0.001) when compared with the diabetic controls (63%). Significant increases in the weight of granulation tissue (p < 0.001) and hydroxyproline content (p < 0.00 1, 92.16 +/- 4.02) were observed. The protein content was moderately high. Histological studies showed that collagen was laid down faster in the experimental diabetic animals than in the normal control and diabetic control groups. Fasting blood glucose values in the diabetic experimental group had reduced by 29% (p < 0.00 1) compared with the diabetic control animals. There was a good correlation between the wound contraction rate and blood glucose values. CONCLUSION: This study demonstrates that the juice of Morinda citrifolia fruit significantly reduces blood sugar levels and hastens wound healing in diabetic rats.

An incidence of canine circumanal gland adenoma (CGA) is reported, apparently for the first time, in the island of Trinidad and Tobago. The case involved an 11-year-old Rottweiler dog which was presented in relatively good health except for noticeable masses around the perianal region. Apart from routine histopathological investigation which enabled a definitive diagnosis of CGA with secondary vascularization, analysis of the nuclear chromatin texture was performed on the digitalized images of both reserve and hepatoid cells of the neoplasm. The mean nuclear pixel intensity value of the images of processed CGA reserve cells was significantly lower than those of the processed CGA hepatoid cells (P<0.1). Our observation based on the texture analytical data suggests a high degree of rate of multiplication of the reserve cells at the expense of degenerating hepatoid cells. The relevance of this finding is discussed within the context of vascular supply to the neoplastic cells (AU)

The efficacy of Lamstreptocide A & B was studied on 9 natural cases of bovine and caprine dermatophilosis in 8 different farms in St. Kitts, employing standard histopathologic and bacteriological methods. The lesions of 5 of the treated cases were dried-up, and there was marked peeling-off of scabs of a severely affected case exposing erythematous underlying tissue, at 3 weeks post-application of the product. Apart from 3 mild cases which were not available for follow-up studies and which were reported to have recovered, there was no outright recovery of the 5 animals after treatment at 3 weeks, and even after a second application of the product. An in vitro sensitivity test of the product revealed a slowing down of growth of Dermatophilus congolensis at concentrations in excess of 1% by agar-streak method. However, there was no inhibition of growth of the bacterium by an agar-impregnated sensitivity method.

By combined electron microscopical and serological methods the rate of involvement of pox virions within lesions of bovine natural dermatophilosis was determined. Scabby lesions of 40 (77%) out of a total of 52 cases were positive for the virions, while sera of all infected animals which reacted positively for pox viral antibodies (LSD) was significantly higher (P less than 0.001) in comparison to those of healthy-appearing animals. The virions in all cases had a uniform oval to cylindrical morphology with a mean measurement of 110 X 280 nm. A synergistic interplay involving the initiation of the scabby lesions by the virions and their subsequent trapping within keratin deposits, followed by their release through the keratolytic activity of Dermatophilus congolensis appear to have obtained in cases.

Naturally occurring canine transmissible venereal tumors in 2 dogs were examined ultrastructurally, using a transmission electron microscope. Typical nondegenerating cells of various shapes with normal cytoplasmic organellae were seen. Degenerating and necrotic cells characterized by loss of normal organellae with vacuolations of the cytoplasm were in all sections studied. Icosahedral electron-dense particles (22 nm diameter) were in the degenerating and necrotic cells. The uniqueness of this finding is not known. The shape and size of the particles indicate that there may be viral involvement.

RESUMO

Transmission electron and light microscopy studies were carried out on morphologic characteristics of cellular organelles of normal and different neoplastic cells of hepatoid circumanal glands of 16 dogs. The ultrastructural features of the reserve and transitional cells in the adenomas and carcinomas are highly suggestive of different carcinogenic stimuli for each form of hepatoid circumanal gland neoplasm. The reserve cells, in response to the carcinogenic stimuli, appear to be growing at the expense of the degenerating glandular cells.

RESUMO

Intranuclear cisternal and vesicular structures, bound by a single membrane, were observed for the first time in some glandular "bird's eye" cells of canine circummanal gland carcinomas. The role of these intranuclear structures is not clear. It is suggested that they may be involved in cellular adaptation to an environment of nutrient deficiency, as a result of cell overcrowding.
